This kit employs a sandwich enzyme immunoassay technique to quantify VILIP-1 (VSNL1) in biological samples.

Introduction

VSNL1 encodes VILIP-1 (visinin-like protein 1), a small calcium-binding protein belonging to the neuronal calcium sensor family that is related to the visual protein recoverin. VILIP-1 is particularly abundant in cerebellar granule neurons, where rising calcium levels cause it to associate with the cell membrane and, from there, adjust the activity of downstream signaling enzymes that shape neuronal excitability and intracellular messaging. Reported binding partners extend beyond a single pathway and include specific guanylyl cyclases and a subtype of nicotinic acetylcholine receptor, along with an ATP-gated ion channel, consistent with a broader role as a calcium-dependent switch that fine-tunes several signaling routes rather than just one. Because neuronal injury or degeneration can release VILIP-1 into cerebrospinal fluid and blood, its levels have drawn interest as a potential indicator of neuronal damage in conditions such as Alzheimer’s disease and epilepsy.

Principle of the Assay

This kit uses a sandwich enzyme immunoassay format to detect and quantify Human VILIP-1 (VSNL1). The microtiter plate wells are pre-coated with a VSNL1-specific capture antibody that binds the target protein present in standards or samples. A biotin-conjugated detection antibody, also specific to VSNL1, is then added to form a capture antibody–analyte–detection antibody sandwich complex. Avidin conjugated to horseradish peroxidase (HRP) is subsequently added to each well, where it binds the biotin moiety of the detection antibody. Upon addition of TMB substrate solution, HRP drives a colorimetric reaction exclusively in wells containing the assembled complex. The reaction is stopped by the addition of a sulphuric acid stop solution, and absorbance is measured spectrophotometrically at 450 nm (± 10 nm). Sample VSNL1 concentrations are then determined by referencing the measured optical densities against a standard curve.
